These supplies you give him carry over across all your runs through the Gungeon so you're not expected to give him everything at once. As he repairs, you need to bring him supplies. The Tinker will be waiting for you in the elevator shaft of whichever floor he's repairing. Once he has completed repairs there he will go down to Chamber 3, and so on until you're able to access any of the 5 main chambers at will. Clear the first Chamber, the Keep of the Lead Lord, and descend down to Chamber 2, the Gungeon Proper. The Tinker isn't very difficult to locate.
